Basically, you submit a design that you think would look cool on a t-shirt, and the community critiques and rates your design. The best designs win $2,000, along with a $500 Threadless gift card. (I think you can redeem it for $400)
You also get an additional $500 for each reprint. So, if you need cash and you don't care about getting screwed, (As they probably make waaay more money off your design than you do) then try it out.

You can also submit your witty slogans to TypeTees, for a chance at $500.
